Malhas de polipropileno recobertas com colágeno polianiônico ou como dupla camada com poli(cloreto de vinila) para a reconstrução da parede abdominal

Resumo

Malhas poliméricas são, atualmente, os materiais mais in dicados para a reconstrução da parede abdominal, com destaque para as malhas de polipropileno. Os objetivos deste trabalho foram desenvolver, caracterizar e avaliar o comportamento biológico de malhas de polipropileno recobertas com colágeno polianiônico ou na forma de du pla camada com poli(cloreto de viníla), como biomateriais para a reconstrução da parede abdominal. Os materiais foram caracterizados por eletroforese em gel de poliacri lamida-SDS, espectroscopia no infravermelho, calorimetria diferencial de varredura, microscopia eletrônica de varre dura e por implantes na parede abdominal de carneiros por períodos entre 1 e 18 semanas. A malha de polipro pileno, na forma de dupla camada com poli(cloreto de viníla), mostrou-se mais biocompatível e menos susceptí vel à adesão das alças intestinais, sugerindo seu potencial para uso na reconstrução da parede abdominal. Os resul tados mostraram também que o poli(cloreto de viníla) isoladamente e, principalmente, pela ausência de adesão após 9 semanas, pode constituir-se em um biomaterial para uso temporário e de baixo custo. Embora a neoformação óssea observada com os implantes de polipropileno : colágeno polianiônico não recomende o seu uso como o proposto inicialmente neste trabalho, este fato constitui-se em uma indicação de uma ação ósteo-indutora do colágeno utili zado, uma propriedade importante para materiais dirigi dos à reconstrução de tecido ósseo.

Abstract

Polymeric mesh are the most recommended materials for the reconstruction of the abdominal wall, with particular emphasis for polypropylene mesh. This work describes the preparation, characterization, and the biocompatibility studies on polypropylene mesh coated with polyanionic collagen or in the form of a double layer with poly(vinyl chloride), intended as a biomaterial for the reconstruction of the abdominal wall. Materials were characterized by SDS-polyacrylamide gel electrophoresis, infrared spectroscopy, differential scanning calorimetry, electron scanning microscopy, and by implantation in the abdominal wall of sheep for periods from 1 to 18 weeks. The results showed that the double layer of polypropyleno : poly(vinyl chloride) was the most biocompatible, and the list susceptible with respect to bowel adhesion, suggesting its potential use as a temporary or permanent biomaterial for abdominal wall reconstruction. For the same reasons, poly(vinyl chloride) may constitute in an efficient and low cost biomaterial for short term abdominal wall repair, particularly due to the lack of bowel adhesion after 9 weeks of implantation. In the case of polypropylene mesh coated with polyanionic collagen, although the formation of bone tissue around the implant does not recommend its use for the purpose intended in this work, the results suggest that polyanionic collagen may be an osseo-inducting biomaterial, a very useful property for biomaterials intended for the reconstruction of bone tissue.
